mRN4 in human hepatoma cell lines

چکیده
A solution hybridization/RNase protection assay with riboprobes was developed to quantitate apolipoprotein mRNA concentrations. Previously, radiolabeled DNA probes have been used in solution hybridization/Sl nuclease protection assays for this purpose. The new assay requires less time for probe preparation and hybridization compared to previous assays. In addition, the vector used for riboprobe preparation can also be used to conveniently produce cRNA required to generate the standard curve to quantitate absolute apolipoprotein mRNA levels. The solution hybridization RNase protection assay was used to quantitate apoB, A-I, and E mRNA levels in four human hepatoma cell lines, HepGP, HepJB, WRL-68, SK-HepP. HepG2 and Hep3B, but not WRL-68 and SK-Hep2 cells had concentrations of all three apolipoprotein mRNAs comparable to liver in vivo. These data suggest that HepGP and Hep3B are suitable models to study liver specific apolipoprotein gene expression. -Azrolan, N., and J. L. Breslow. Gene Expression under F8 Promoter Driving In Mouse Hepatoma Cells: A Step towards Gene Therapy of Hemophilia
Background and Objectives: Significant progress has been made in treatment of hemophilia. Ex-vivo gene therapy is going popular due to the capability of this method in using isogenic cells for genetic manipulation and reintroducing them into same host after proliferation. Most gene therapy techniques use viral vectors, which usually harbor a strong and non-specific promoter (e...

Introduction & Objective: Hepatocellular carcinoma (HCC, also called malignant hepatoma) is a primary malignancy (cancer) of the liver. Most cases of HCC are secondary to either a viral hepatitic infection (hepatitis B or C) or cirrhosis (alcoholism being the most common cause of hepatic cirrhosis. Recently, the biological activities of zingiber officieale plants have been reported as possess...
متن کاملAntioxidant Effects of Pinus Eldarica Extracts Against Cisplatin-Induced Cytotoxicity in Human Hepatoma (Hepg2) Cell Line
Background: Cisplatin has cytotoxic effects through free radical generation. Reports indicate that various members of Pinaceae family may have antioxidant properties. In this study we investigated the cytoprotective effects of needle volatile oil and bark extract of Pinus eldarica in hepatoma G2 (HepG2) cell line. Methods: Using the maceration technique, we obtained the ethanolic extract of ...
متن کاملCytotoxicity of Subtoxic AgNP in Human Hepatoma Cell Line (HepG2) after Long-Term Exposure
Background: We aimed at evaluating the toxicity effects of low (subtoxic) concentrations of silver nanoparticles (AgNP, 5-10 nm) in human hepatoblastoma (HepG2) cell line after and during a period of about one month. Methods: XTT and MTT assays were used to draw a dose-response curve IC50 (half maximal inhibitory concentration) value of the AgNP on HepG2 cells was calculated to be 2.75-3.0 mg/l...
متن کامل - TGF ، سنتز آلفا ـ 1 ـ آنتی تریپسین را در سلولهای هپاتومای انسانی (HepG2)
Cytokines protein or glycoproteins synthesized by a wide range of cell types including lymphocytes, macrophages, platelets, and monocytes. Cytokines induse synthesis of acute phase protein in hepatocytes. So far eight cytokines has been shown can induce acute phase protein synthesis in hepatocyte and hepatoma cell culture. There are evidences that transforming growth factor-alpha (TGF-a) secr...
